Improvement of β-mannanase production by some fungi and its application
β-mannanase, Penicillium citrinium, Gamma, Immobilization and Nematode
The aim of our study is to improve the biosynthesis of β-mannanase by different techniques as mutation and optimization of the culture parameters. In our study we found stable mutant strain obtained by using gamma radiation at 150 GY gave 37.42 IU/ml while the wild gave 16.48 IU/ml. The first step of the fermentation optimized by one factor at time technique increased the biosynthesis of β-mannanase for P. citrinium 150 GY from 37.42 IU/ml to 219 IU/ml comparing to the wild strain that increased from 16.82IU/ml to 26.5IU/ml. Statistical optimization improve P. citrinium 150 GY β-mannanase from 219 IU/ml to 296 IU/ml and using Plackett-Burman design increased to 351 IU/ml using CCD. Primary characterization of β-mannanase produced by P. citrinium and P. citrinium 150 GY proved that they almost the same except in a little shift to higher value (5°C) in optimumtemperature. Fraction precipitation with 60-70% ammonium sulfate represented 34.03% of the total recovered activity with 7.99 times increase in the specific activity.
