PHYTOCHEMICAL AND BIOLOGICAL EVALUATION OF PLEIOGYNIUM TIMORENSE (DC.) LEENH.(FAMILY: ANACARDIACEAE) GROWING IN EGYPT

pleiogtnium timorense (DC.) Leenh. (Anacardiaceae)' is an evergreen tree native to Australia. The plant has many Synonyms as Pleioglniu:l solandti and Pleiogynium cerasiferum.ItisknowninArabicasGambozia,andiscultivatedinEgyptaSan omamental plant . It is one of the plants having edible fnrits used in preparation of jelliesandjams.Thepresentworkincludesphytochemicalscreeningwhich revealed the presence of flavonoids, coumarins, sterols andlor triterpenes, carbohydratesand/orglycosidesandtanninsinpericarpandseedsoftheplant. GC/I\4S analysis of of unsaponifiable matter (usM) from the petroleum ether extract of Pleiogynium timorense seeds revealed 28 compcunds (hydrocarbons and sterols), representing 96.65% of the total unsaporiifia'ole c6ntent' l-Heptene (66.47%)isthemajorcompoundintheUSM,followedbyButylatedhydroxy toluene Ql.o1%). Two steroidal compounds were identified ; Cholesterol, BSitosterol and one triterpenoidal compound was identified ; o-amyrin' GC/MS analysis of the fatty acids fraction as methyl ester derivatives revealed the presence of g fatty acids, representin g95.84%of the total composition' Major fatty acids are9,12-o ctadecadienoic acid (linoleic acid) (33 .8%),followed by 9-octadecenoic acid (oleicacid)(24.14%).Studyoftheconstituentsfromthepetroleumetherextractof the seeds of Pleiog,,nium timorense resulted in the isolarion and identification of twocompounds:CompoundLtwasidentifiedaso_Amyrinwhichwasisolated beforefromtheleavesoftheplant.CompoundL2wasidentifiedas5,24(28)- cholestadie n-24-met$len-3beta-o1 isolated for the first time from the plant' Methanol extracts of defatted powder of Pleiogtnium timorense pericarp and seeds were subjected to HPLCiMS analysis and tandem MSa{s' Twenty five and twenty seven compounds were identified from Pleiogtnium timorense pericarp and seeds ,respectivlyDifferentphenolicacidsandflavonoidswereidentified.Six compounds were isolated from the pericarp of P'timorense' whichwere identified asrutiir,quercitrin,quercetin,gallicacid,pinobanksinandcatechinFour ,rll.compounds were isolated {iom the seeds of P.timorense' which were identified as catechin ,para methoxy benzaldehyde, pyrogallol and gallic acid 'From the seeds of the plant, sulphur -containing compound (sulpholane) was isolated for the first time from the plant and from the family. Determination of LDso of methanol (70%) extracts of the pericarp and the seeds of P.timorense indicates the low toxicity of th. both plant. Serum glucose level was significantly decreased by the treatment with pericarp and seed extracts. . Both seed and pericarp extracts showed significant antioxidant, hepatoprotective and renal function protective avtivities, while pericarp extract was more powerful than that of seeds. Both seed and pericarp extracts showed significant analgesic activity' Pericarp extract showed significant antiinflammatory effect. More than that of the seed extract'The methanolic extract (70%) of P.timorense pericarp showed moderate cytotoxic activity against the liver cancer cell line, while that of the seeds showed weak cytotoxic activity against the used human tumor cell lines. Methanol extracts (70%) of P. timorense peticarp and seeds have weak antiviral activity against Avian Influenza A Virus (H5N1) incomparison to reference drug Zanamivir'
