controlled release (cR) dosage forms offer a lot of advantages such as; patient convenience, reduction of dosing frequency and improved safety. In addition, cR dosage forms are considered more economic as they enable reduction of the totar amount of drug administered. Gliclazide is a second generation surphonyl,rea, oral antidiabetic drug. It exhibits low solubility and high permeability (Biophannaceuticar classification system, Bcs, class If). The low water-sorubitity of gliclazide leads to a slow dissolution rate. Therefore, dissolution is considered as the rate limiting step for gricrazide absorption. It exhibits high inter-subject variation in its absorption. oral administration of a single dose of glicrazide results in a varying maximum prasma concentration that ranges from 2.2 to g mg/L within 2 to g hours. This variability in the absorption was attributed to both the physicochemical properties of gliclazide as well as the formulation administered. The recommended griclazide dose is much lower in case of modified release dosage forms compared to the immediate release ones. Hence, it would be more economic and beneficial to formulate a cR drug deriverysystem.
