Cancer is a major cause of mortality however advances in surgery and therapy. Plants such as Annona muricata have played in the treatment of cancer and infectious diseases. Screening of the endophytic fungi associated with Annona plant parts indicated that a total of sixty five endophytic fungal isolates were isolated. Molecular identification done for A. niger isolate (2), the most frequent species, revealed that it is A. niger strain SH3 using 18S rDNA and phylogenetic analysis.It has accession number of MG847141.1 in (https://www.ncbi.nlm.nih.gov/nuccore/MG847141)GenBank. Phytochemical analysis were tested for different Annona parts along with the isolated endophytic fungi and indicated the occurrence of alkaloids, flavonoids, terpenoids, phenols, proteins and anthraquinone. In vitro cytotoxicity assay of the endophytic fungal extracts in addition to seed, leaves and fruit extracts was done against human cell lines of breast cancer (MDA-MB-231), cell lines of colon cancer (Caco2) and liver cancer (HepG-2) using MTT assay method and indicated the effectiveness of R. solani, P. jensenii, A. niger, A. niger Strain SH3 and seed extracts specifically against Caco2 cancer cell line. Gas Chromatography Mass Spectroscopy (GCMS) was used for analysis of extracts which showed the most cytotoxic activity in In vitro study which showed that alkaloids, terpenoids and phenolic compounds are dominant in fungal extracts while fatty acids are dominant in Annona seed extract . Combination study has been made between A. niger strain SH3 and seed extracts against against human cell lines of colon cancer (Caco2) and liver cancer (HepG-2).
