This work aims to isolate bacteria capable of producing α-amylase enzyme and identify them via the morphological, physiological and molecular characteristics.  PCR sequencing of the gene coding for α-amylase enzyme isolated from the most potent strain.  Increasing the enzyme production using one-factor-at-a-time and statistical methodology (Plackett-Burman followed by Central Composite design).  Covalent immobilization of α-amylase on different carriers and selecting the immobilized enzyme with highest immobilization yield (IY %) and immobilization efficiency (IE %).Optimization the modification of Chitosan-MNP/PEI/GA beads using Central Composite design to promote IY%.  Characterization of some parameters affecting free and immobilized α-amylase activity and stability.  Comparing the catalytic, kinetics and thermodynamics of both free and immobilized α-amylase for evaluating its suitability for industrial applications
