GENETIC AND PHYSIOLOGICAL EVALUATION OF SOME SUGARCANE (SACCHARUM OFFICINARUM) CULTIVARS

This study includes application of in vitro propagation, molecular markers and finally biochemical analysis of invitro propagation on two sugarcane cultivars (Saccharum officinarum L.).The obtained results revealed that, an efficient in vitro propagation method developed using fresh apical meristem. Explants cultured on MS medium supplemented with 1mg/L BAp+ 0.25 mg/L NAA resulted, highest rate of shoots multiplication with GT54-C9 cultivar. While, for G.2003/49 cultivar highest rate of multiplication was recorded with MS medium supplemented with 2mg/L Kin+0.25 mg/L NAA. Shootlets were rooted on half salts strength of MS medium supplemented with 1mg/L IBA +1mg/L NAA for GT54-C9 cultivar. While, for G.2003/49 cultivar shootlets were rooted on half MS medium supplemented with 1mg/L NAA. In vitro plantlets were transplanted to greenhouse for acclimation and adaption which resulted in survival percentage of 80-90% in both cultivars. Concerning the biochemical analysis of soluble carbohydrates, total carbohydrates; flavonoid, phenol, indols and chlorophyll contents (Carot, ChI b, and ChI a) plant growing in open field (as a control), regenerated plantlets and acclimatized plants for each cultivar. The DNA finger prints markers (ISSR- PCR) resulted in highly similarity between them. Keywords: Saccharum
