Large scale screening of selected Egyptian plant extract's against human colon, liver, breast and lung carcinoma cell lines for detection of antitumor activity in-vitro

Forteen plants belonging to 6 families were collected from different areas in Egypt. These plants yielded 50 methanol extracts and were tested for their anticancer, free radical scavenging and anticandidal activities. The anticancer activity was tested against four human cancer cell lines, namely, A549 (lung carcinoma), HCT-116 (colorectal carcinoma), HepG2 (hepatocellular carcinoma) and MCF-7 (breast adenocarcinoma). Cell viability was determined using the MTT (3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyltetrazolium bromide) assay. Free radical scavenging activity was tested using 2, 2 diphenyl-1-picrylhydrazyl (DPPH) assay. Anticandidal activity was measured using disk diffusion method.Results showed 11 methanol plant extracts possessed both high cytotoxicity and free radical scavenging activities, these included: branch extract of Anacardium occidentale,bark extract of Pleiogynum solandri, branch extract of Pistachia terebinthus, leaves extract of Harpophyllum caffrum, leaves extract of Albizzia stipulate, branch extract of Derris robusta, bark extract of Bauhinia variegate, bark extract of Adenanthera pavonina, branch of Albizzia stipulate, leaves of Derris elliptica and bark of Hyphaene thebaica. On the other hand, 14 methanol plant extracts possessed high free radical scavenging activity and showed weak cytotoxicity against the tested cancer cell lines. All these promising extracts showed no activity against candida albicans except the branch extract of Albizzia stipulate which possessed moderate anticandidal activities. Leaves and fruit extract of Aegle marmelos showed anticandidal activity but did not possess neither cytotoxic nor free radical scavenging activities. These promising extracts were further tested for cytotoxicity on human normal cell line (BJ-1) to calculate their selectivity index (SI) values. In vitro cytotoxic effect on 3D cancer spheroids were carried out for the most active plant extracts to select the most active tobe subjected to initial fractionation. Six extracts showed moderate to high cytotoxic effect on 3D cancer cells: the leaves extract of Albizzia stipulate, bark extract of Hyphaene thebaica, leaves extract of Harpophyllum caffrum, branch extract of Pistachia terebinthus and Albizzia stipulate and leaves extract of Derris elliptica. These extracts are considered the most promising for anticancer activity in the context of this study and deserve further chemical and biological investigations.
