Genetic polymorphism of some multi-functional genes in Egyptian buffalo
buffaloes / PCR-RFLP / GH/Alu, GHR/Alu / GHRH/GHRHR / IGF
The alleles and genotype frequencies of the GH/Alu, GHR/Alu, GHRH/HeaIII, GHRHR/Eco571, IGF-1/SnaB and IGF-1R/Taq1, gene polymorphisms in Egyptian buffaloes (Bubalus-bubalis L) were estimated using PCR-RFLP technique. Genomic DNA were extracted from blood samples of healthy buffaloes. They were amplified using primers that were designed from the cattle genes sequences and digested with the specific restriction enzymes. The amplified fragments obtained were found to be 211, 342 and 296 bp with only one SNP (A/C), (T/A) and (T/C), and genotyped as LL, GG and AA for GH, GHR and GHRH respectively. The provided amplicon for GHRHR was found to be 425 without any SNP, and genotyped as AA. Whereas the amplified fragments of IGF-1 gene was found to be 250 bp with one SNP (T/A). The current samples were genotyped as BB. The length of the amplicon of IGF-1R was 616 bp and 13 SNPs wereobserved at different positions. The examined animals were genotyped as AA. The nucleotide sequence data were submitted to NCBI /Bankit/GenBank with certain accession numbers. GH: JN809916, GHR: JN809917, GHRH: JN967799, GHRHR: KC295414, IGF-I: KC852883 and IGF-IR: KV852884 for Egyptian buffalo. The study of variation in these sequences may be useful in future genotype-assisted selection (GAS) andgenetic conservation programs.
