Assessment Of Nutritional And Bioactive Constituents From Some Microalgae (Dunaliella Salina, Scenedesmus Obliquus And Spirulina Platensis)
Microalgae, Dunaliella salina, Scenedesmus obliquus , Spirulina platensis, lipids, carotenoids, protein, polysaccharides, Hepatitis-C virus, Rotavirus, Herpes simplex virus, Coxsackievirus, hepatocellular carcinoma (HepG2), breast adenocarcinoma (MCF7) , colon carcinoma (HCT116), antioxidant and anti-cholinesterase.
Laboratory and mass production of Dunaliella salina (D. salina), Scenedesmus obliquus (S. obliquus) and Spirulina platensis (S. platensis) were performed in the algal biotechnology unit, National research centre. The produced three microalgae were subjected to phytochemical investigation of the lipoidal matter, carotenoids, protein and polysaccharides contents and in-vitro biological studies including cytotoxic activity against HepG2, MCF7 and HCT116 cell lines as well as antiviral activity against HCV, rotavirus, Herpes simplex virus and coxsakievirus. Furthermore, in-vitro DPPH scavenging and anticholinesterase activities were studied. Results of the lipoidal matter study revealed highly significant cytotoxic (against MCF7 and HepG2), antioxidant and anticholinesterase activities of the pet.ether extract of D. salina. Also, highly significant cytotoxic (against MCF7) and anticholinesterase activities of the chloroform extract of D. salina were observed. With respect to S. obliquus, campesterol, β-sitosterol, cholesterol, palmitic acid and oleic acid were detected. Promising highly significant cytotoxic (against MCF7, HCT116 and HePG2) and significant antioxidant activities of the pet.ether extract of S. obliquus were detected. Also, highly significant antioxidant and significant anticholinesterase activities of the chloroform extract of S. obliquus were observed. Concerning S. platensis, chondrillasterol (phytosterol), PUSFs, omega 3 and omega 6 fatty acids were detected. significant antioxidant and good antiviral activities of the pet.ether extract of S. platensis were detected. Also, highly significant cytotoxic (against MCF7) and highly significant antioxidant and activities of the chloroform extract of S. platensis were observed. Results of the carotenoids study revealed the detection and identification of six carotenoids using ESI+-MS in S. platensis and S. obliquus while four carotenoids were identified in D. salina. S. platensis and D. salina carotenoids showed significant inhibition against liver, breast and colon cancer cell lines. Also, significant antioxidant activity while D. salina had the highest promising anticholinesterase activity. Moreover, The proteins of the three microalgae were isolated in which polyacrilimide gel electrophoresis revealed the presence of low molecular weight peptides. Also, amino acid analysis revealed the presence of essential and non-essential amino acids. D. salina protein exhibit promising highly significant antioxidant activity. Also, D. salina and S. obliquus exhibit moderate anticholinesterase activity. Polysaccharides study revealed that S. platensis had promising antiviral activity against all the tested viruses while D. salina hot water polysaccharides HEM show potent cytotoxic activity against HCT 116. Also, D. salina HEM and S. obliquus cold water extracted polysaccharides CEM showed high significantly antioxidant activity. and S. obliquus CEM and HEM exhibited high significant anticholinestearse activity.
